Introduction

Microtubules constitute one of the major components of eukaryotic cell cytoskeleton and are involved in many essential processes, including cell division, ciliary and flagellar motility and intracellular transport. Microtubules of the eukaryotic cytoskeleton are composed of a heterodimer of alpha- and beta-tubulin. In addition to alpha- and beta-tubulin, several other tubulins have been identified.

Alpha and beta-tubulins alternate with one another in 13 linear filaments to form microtubules. Microtubules serve as structural supports and lines of transport within the cell, as well as serving a key role in mitosis. The function of microtubules is dependent on their ability to grow and shrink. This dynamic instability is dependent on many factors, the main one being the concentration of GTP-bound tubulin heterodimers. Also, molecules called microtubule-associated proteins (MAPs) can inhibit or stimulate the polymerization of microtubules.

Class III beta tubulin is a vertebrate tubulin isotype only found in neurons and cells in the mammalian testis. Therefore, Class III beta tubulin antibodies are commonly used as neuronal markers.

Gamma-tubulin is found at microtubule organizing centers (MTOC) such as the spindle poles or the centrosome, suggesting that it is involved in the minus-end nucleation of microtubule assembly.

Delta-tubulin function is an essential component during spermatogenesis.

Epsilon-Tubulin was found to be located in the centriolar area, a localization that is dependent on cell cycle modulation.

Tubulins are generally highly conserved proteins throughout all eukaryote species. Therefore, most available antibodies to tubulins are reactive on nearly all species tested so far.
