Endothelial Differentiation Gene Receptors
(EDG-1,-2,-4,-5,-6,-7 and -8)
Introduction
Lysophosphatidic acid (LPA), sphingosine 1-phosphate (S1P) and some other structurally related lysophospholipids are active growth factors and stimuli for diverse cellular functions.
LPA and S1P promote early T cell migration to tissue sites of immune responses and regulate T cell proliferation and secretion of numerous cytokines. The endothelial differentiation gene (EDG) receptors are a class of G protein-coupled receptors. EDG-1, -3, -5, -6, and -8 (known as the endothelial differentiaton gene-1 family) regulate diverse processes including cell migration, angiogenesis, vascular maturation, heart development, neurite retraction and soma rounding. These receptors bind the bioactive lipid sphingosine-1-phosphate (S1P) as the primary signaling ligand. EDG-2, -4, and -7 bind the ligand lysophosphatidic acid.
