Antibodies to BAI1,2,3 and BAIAP2
Introduction
Angiogenesis is controlled by a local balance between stimulators and inhibitors of new vessel growth and is suppressed under normal physiologic conditions. Angiogenesis has been shown to be essential for growth and metastasis of solid tumors. In order to obtain blood supply for their growth, tumor cells are potently angiogenic and attract new vessels as results of increased secretion of inducers and decreased production of endogenous negative regulators. It is reported that the genetic alteration of p53 is associated with neovascularization during the progression of glioma to its more malignant form, glioblastoma. Brain-specific Angiogenesis Inhibitor 1 (BAI1) which was described as a p53-target gene, is specifically expressed in the brain, however is not expressed in gioblastoma cell lines, suggesting that BAI1 plays a significant role as an inhibitor of angiogenesis and a growth suppressor of glioblastomas. The finding that expression of BAI proteins may be lost during tumor formation is of special interest as restoration of their function in tumors may be of therapeutic benefit.
There are two other brain-specific angiogenesis inhibitor genes, designated BAI2 and BAI3 which along with BAI1 have similar tissue specificities and structures. The expression and distribution of BAI3 in normal brain, but not its developmental expression, seem to be very similar to those of BAI1 and BAI2, and BAI3 may participate in the early phases of ischemia-induced brain angiogenesis and in brain tumor progression.
BAI1-associated protein 2 (BAIAP2) has been identified as a brain-specific angiogenesis inhibitor (BAI1)-binding protein. This interaction at the cytoplasmic membrane is crucial to the function of this protein, which may be involved in neuronal growth-cone guidance. This protein functions as an insulin receptor tyrosine kinase substrate and suggests a role for insulin in the central nervous system. It has also been identified as interacting with the dentatorubral-pallidoluysian atrophy gene, which is associated with an autosomal dominant neurodegenerative disease. It also associates with a downstream effector of Rho small G proteins, which is associated with the formation of stress fibers and cytokinesis.
Antibody Panel to BAI1,2,3 and BAIAP2
Acris Antibodies offers a range of polyclonal antibodies to BAI1, 2 and 3 and BAI1-associated binding protein 2 (BAIAP2). The antibodies to inhibitor types 1, 2 and 3 have been produced to different epitopes and can be used for immunohistochemistry on paraffin embedded sections. Antibody SP7258P to BAIAP2 is suitable for western blot application. The antibodies stain human and mouse samples; the antibody to BAIAP2 also shows rat cross reaction (see pictures and table of antibody panel below).
