| Background | NF-κB is a family of transcription factors, including p50, p52, p65, RelB, and C-Rel (1). It regulates the expression of a number of proteins involved in the regulation of cell survival and immune responses (2). The biological activity of NF-κB is tightly controlled by its inhibitor protein, IκB, which binds to NF-κB and sequesters it in the cytoplasm. There are three isoforms of the IκB: IκBα, -β and â??ε. IκBα, the well-characterized member of the IκB family contains both nuclear localization sequence and nuclear export signal that render to IκBα the activity to rapidly shuttle between the nucleus and the cytoplasm. The IκBα protein binds to NF-κB dimmers, masking their nuclear localization sequences and causing their cytoplasmic retention. After NF-κB positive stimuli, IκB kinase complex phosphorylates IκBs in two conserved serines (Ser-32 and Ser-36 in IκBα), leading to ubiquitination and subsequent proteasomal degradation (1). |

| Specificity | This antibody is specific for IκBα and is phosphorylated at the position of serines 32 and 36. The antibody was evaluated by ELISA, dot blot and Western blot. It only recognizes the phosphorylated serines 32 and 36 of IκB, not other phosphorylated sites or non-phosphorylated peptides. By immunoblot assay, this antibody recognizes ~40 kDa band in the TNF-treated Hela or 3T3 whole cell lysates. This immunoreactive band is abolished upon addition of the immunizing peptide. Species: Human, Mouse, Rat. |
