| Background | c-Raf, also known as Raf-1, a member of the MEK kinase family, is a 74 kDa serine/threonine kinase which is activated in response to a variety of mitogens and cytokines. Activation of Raf kinase activity is induced by several events including phosphorylation at several sites and binding of GTP-bound Ras and/or 14-3-3 proteins. This, in turn, leads to the phosphorylation and activation of other members of the MEK/MAPK/ERK cascade. Phosphorylation of tyrosines at position 340 and 341 iscatalyzed by the Src tyrosine kinase and this event is involved in c-Raf activation. |
