BackgroundF4/80 antigen is a member of the EGF-TM7 family of proteins which shares 68% overall amino acid identity with human EGF module-containing mucinlike hormone receptor 1 (EMR1). Expression of F4/80 is heterogeneous and is reported to vary during macrophage maturation and activation. The F4/80 antigen is expressed on a wide range of mature tissue macrophages including Kupffer cells, Langerhans, microglia, macrophages located in the gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma and macrophages in the red pulp of the spleen. F4/80 expression has also been reported on a subpopulation of dendritic cells but is absent from macrophages located in T cell areas of the spleen and lymph node.
The ligands and biological functions of the F4/80 antigen have not yet been determined but recent studies suggest a role for F4/80 in the generation of efferent CD8+ve regulatory T cells.

ApplicationsFlow cytometry: Use 10 µl of neat to 1/10 diluted antibody to stain 10e6 cells in 100 µl.
Immunohistology on frozen and paraffin embedded sections. For paraformaldehyde paraffin tissue, pretreatment by trypsinisation for 20-30 minutes is required. Use 1:50 on cryosections, 1:20 on paraffin sections.
Western Blot.
DescriptionCI:A3-1 recognizes the F4/80 antigen, a 120-160kD glycoprotein containing 7 EGF-like domains at the N-terminus, an RGD (Arg-Gly-Asp) integrin binding motif, and significant homology to the transmembrane 7 (Tm7) hormone receptor family at the C-terminus. The molecular structure suggests multiple ligands. Possibly, F4/80 is involved in macrophage adhesion within certain tissues, combined with receptor signaling following peptide-ligand interaction. The antigen is expressed by most macrophages and macrophage precursors beyond M-CFC, and increases upon maturation. Activated macrophages, and eosinophils express low levels. Freshly isolated spleen dendritic cells are uniformly positive for F4/80, but the expression decreases upon culture. This antibody is very useful for characterising tissue fixed macrophages, particularly in combination with BM 8 and MOMA-2.
SpecificityBM4008B recognizes the murine F4/80 antigen, a 160 kD cell surface glycoprotein.
