BackgroundMultidrug resistance (MDR) is a phenomenon that renders cells resistant to chemotherapy in most cancer patients. One of the mechanisms that is responsible for this pleiotropic effect is a reduced drug accumulation inside the cells, due to the activity of a transmembrane protein, the P-glycoprotein (P-gp; other designations P-170, mdr1 gene product) with a molecular weight of 170 kDa.
The P-glycoprotein is assumed to act as an ATP-dependent drug efflux pump with broad substrate specificity.
Tissue Distribution: MDR mechanisms exists in numerous healthy cells and protects them from ''naturally occurring'' drugs such as anthracyclines, vinca alkaloids, podophyllotoxins and colchicine.
P-gp is mainly expressed in specialized epithelial cells with secretory or excretory functions. In the liver, P-gp is found on the biliary surface of hepatocytes and small biliary ductules; in the pancreas, on the laminal surface of the epithelial cells of small ductules and, in the kidney, on the brush border of the proximal tubules.
Additional sites of expression include the gastrointestinal tract, endothelial cells of the brain as well as the adrenal glands.
ImmunogenBalb/c 3T3-1000 cells (human MDR1 cDNA transfected Balb/c 3T3 fibroblasts).
Format
State: Liquid purified IgG fraction.
BufferSystem: PBS containing 2 mg/ml BSA as stabilizer and 0.09% Sodium Azide as preservative.
ApplicationsUseful for Immunohistochemical techniques (both frozen and paraffin-embedded sections, 2-10 µg/ml) and Flow Cytometry (use undiluted).
It is also useful for functional experiments since UIC2 inhibits P-gp mediated efflux and potentiates the cytotoxic action of P-gp transported drugs in MDR cells.
SpecificityReacts specifically with an extracellular epitope of the human 170 kDa P-glycoprotein.
StorageStore the antibody undiluted (in the dark) at 2-8°C.
